The Effect of Glucocorticoid the Synthesis of 72 kDa Heat Shock Protein in the Hyperthermic Treatment of U-937 lLymphoma Cell


Abstract
We designed this study to investigate the effect of glucocorticoid pretreatment on the level of heat shock protein in U-937 cell after heat treatment. The heating was carried out at 43¡É for 30 minutes, and 100 nM hydrocortisone and betamethasone
were
pretreated at four different times; at the same time with heating and at 6, 12, 24 hours before heating. The changes of 72 kDa, 72 kDa, 73 kDa heat shock protein systhesis was analyzed at 1, 2, 3, 4, 6, 8, 12 hours after heating by gel
electrophoresis
and Western blotting.
@ES The results were as follows :
@EN 1. There was significant increase of 72 kDa heat shock protein after heating, but no change of 72 kDa heat shock protein after heating, which means 72 kDa heat shock protein synthesis is more thermo-sensitive than 73 kDa heat shock protein.
2. The 72 kDa heat shock protein began to increase in 2-3 hours after heating and it was at its highest level 4-6 hours after heating and maintained the increased level until to 12 hours after heating.
3. There was significant reduction of 72 kDa heat shock protein when pretreated with hydrocortisone compared with the only heat treatment group, and the reduction was most remarkable in 2-6 hours after heating and most remarkable at the same
time
treatment group.
4. There was also significant reduction of 72 kDa heat shock protein when pretreated with betamethasone compared with control group. And the reduction was most remarkable in 3-6 hours after heating and at the same time treatment.
5. Betamethasone was more effective than hydrocortisone in decreasing the 72 kDa heat shock protein synthesis in each group of pretreated time with glucocorticoid, and the difference was most remarkable at 2-4 hours after heating.
Our results revealed that the combination treatment with heating and glucocorticoid was more effective than heat treatment only in reducing the 72 kDa heat shock protein systhesis. And these results could be applied in clinical trial for the more
effective method of hyperthermia in cancer treatment.
